目的研究不同方法提取的多伞阿魏挥发油化学成分,并初步考察其抑制胃癌细胞SGC-7901的活性。方法分别采用水蒸气蒸馏法、溶剂回流提取法、超声提取法、微波萃取法、微波一超声协助萃取法、渗漉提取法和超临界CO2萃取法制备多伞阿魏挥发油,用气相色谱一质谱联用法分析其化学成分;MTT比色法测定不同方法制备的多伞阿魂挥发油对胃癌细胞SGC-7901的生长抑制作用。结果从7种不同方法制备的多伞阿魏挥发油中共鉴定了74个化合物,只有水蒸气蒸馏法和微波萃取法制备的多伞阿魏挥发油在对胃癌细胞SGC-7901具有很强的抑制作用。结论水蒸气蒸馏法和微波萃取法制备的挥发油均具有小分子单萜类化合物和部分苯类化合物可能是抑制胃癌细胞SGC-7901的主要原因。
AIM To investigate chemical constituents of volatile oils extracted from root of Ferula ferulaeoide by different methods and their anti-gastric cancer cell line SGC-7901 activity in vitro. METHODS Seven extrac- tion methods, including steam distillation extraction, solvent reflux extraction, ultrasonic extraction, microwave ex- traction, microwave ultrasonic-assisted extraction, percolation extraction and supercritical fluid CO2 extraction, were employed to prepare essential oil from root of Ferula ferulaeoide. And their chemical constituents were ana- lyzed using GC/MS. The inhibitory effects of obtained volatile oils against the cell proliferation of SGC-7901 cells were determined by MTT colorimetric method. RESULTS Total seventy-four compounds were identified from those essential oils. By seven methods volatile oils from of steam distillation extraction and microwave extraction could significantly inhibit the growth of SGC-7901 cells in a dose-dependent manner (1μg/mL- 1000μg/mL). CONCLUSION monoterpenes and Volatile oils extreacted from steam distillation extraction and microwave extraction possess benzenes which might be against the growth of SGC-7901 cells.
