目的:研究肺靶向姜黄素明胶微球在大鼠组织分布及肺纤维化大鼠靶向治疗中的作用。方法:将大鼠分别尾静脉给予大鼠姜黄素明胶微球和姜黄素注射液,采用HPLC法测定各脏器姜黄素含量,采用DAS软件计算药动学参数,评价其在大鼠体内的组织分布。以博来霉素造肺纤维化大鼠模型,尾静脉给予姜黄素明胶微球、地塞米松、生理盐水,病理切片比较大鼠肺纤维化情况。结果:姜黄素明胶微球主要分布于肺部,具有良好的肺靶向性,连续给药28 d后,姜黄素明胶微球组大鼠抗肺纤维化程度明显好于对照组。结论:姜黄素明胶微球具有良好的肺靶向性,其靶向抗肺纤维化治疗效果较好。
Objective: To research the distribution of Curcumin gelatin microsphere in rats and inhibitory effects of pulmonary fibrosis. Methods: The rats were given Curcumin gelatin microsphere and Curcumin via tail vein, HPLC was used to determine the content of Curcumin in different organs. The pharmacokinetics parameters was calculated by using DAS 2. 0 program,evaluate tissue distribution of Curcumin. Model of pulmonary fibrosis in rats was induced by Bleomycin,Curcumin gelatin microsphere,dexamethasone,physiological saline were given to rats via tail vein,and compared pulmonary fibrosis in rats by observing the pathological section. Results: Curcumin gelatin microsphere main distributed in lung and had the good lung targeting property. After 28 days,the effect of antipulmonary fibrosis in the group of rats given Curcumin gelatin microsphere was significantly better than the control group. Conclusion:Curcumin gelatin microspheres could improve tendency for lung,and help improving its lung targeting therapeutic effect.
