目的 探究输血对黑色素瘤荷瘤小鼠TH1/TH2平衡及其细胞因子表达的影响.方法 构建黑色素瘤小鼠肿瘤模型,设新鲜血组与储存血组(30只/组):经小鼠尾静脉分别输注新鲜全血(0 d)或储存血(15 d)0.2 mL,分别在输血后15、30 min与1、4、8h收集小鼠全血,用微量标本多指标流式蛋白定量技术(CBA)检测各时间点血浆中细胞因子TNF-α、INF-γ 、IL-2、IL-4、IL-10的表达水平;另设未输血组(6只/组)为对照组.结果 1)新鲜血组与对照组比较,输血后4 h TNF-α分泌水平(pg/mL)分别为9.14±0.83 vs 1.72±0.90(P<0.01);储存血组与对照组比较,输血后4 hIL-4和IL-10表达水平(pg/mL)分别为1.55±0.17 vs 0.29±0.08和5.4±1.39vs0.2±0.08(P<0.01).2)储存血组与新鲜血组比较,输血后1 h TNF-α和INF-γ表达水平(pg/mL)分别为3.67±1.76 vs 7.64±1.04(P<0.05)和0.27±0.16 vs0.49±0.18 (P<0.05);输血后4 h IL-2的表达水平(pg/mL)分别为0.29±0.08 vs 0.89±0.21(P<0.01);输血后4 h IL-10的表达水平(pg/mL)分别为5.40±1.39 vs 2.38±0.54 (P<0.01).结论 输血可促进小鼠体内TH1和TH2细胞因子的表达,而相对于输注新鲜血,输注储存血组小鼠体内TH1细胞因子表达有所降低,而TH2细胞因子表达得到进一步促进,致TH1/TH2失衡,平衡向TH2方向偏移.
Objective To investigate the effects of blood transfusion on TH1/TH2 balance and expressions of cytokines in melanoma-bearing mice.Methods The tumor model of melanoma mice was established.The fresh blood group and stored blood group were set up (30 mice/group).Mice were transferred with 0.2 mL fresh blood (D0) or stored blood(D15) via tail vein.The whole blood in mice were collected at 15 minutes,30 minutes,1 hour,4 hours and 8 hours after blood transfusion respectively.The expression levels of cytokines (TNF-α,INF-γ,IL-2,IL-4,IL-10) in plasma at each time point were detected by cytometric beads array (CBA).Control group was also set up without blood transfusion (6 mice/group).Results 1) The secretion levels of TNF-α(pg/mL) were 9.14±0.83 in the fresh group and 1.72±0.90 in the control group respectively at 4 hour after blood transfusion (P〈0.01).The expression levels of IL-4(pg/mL) were1.55±0.17 in the stored group and 0.29±0.08 in the control group respectively at 4 hour after blood transfusion (P〈0.01).The expression levels of IL-10 (pg/mL) were 5.4±1.39 in the stored group and 0.2±0.08 in the control group respectively at 4 hour after blood transfusion (P〈0.01).2) The expression levels of TNF-α(pg/mL) were 3.67± 1.76 in the stored group and 7.64± 1.04 in the fresh group respectively at 1 hour after blood transfusion (P〈0.05).The expression levels of INF-γ(pg/mL) were 0.27±0.16 in the stored group and 0.49±0.18 in the fresh group respectively at 1 hour after blood transfusion (P〈0.05).The expression levels of IL-2 (pg/mL) were 0.29±0.08 in the stored group and 0.89±0.21 in the fresh group respectively at 4 hours after blood transfusion (P〈0.01).The expression levels of IL-10 (pg/mL) were 5.40± 1.39 in the stored group and 2.38±0.54 in the fresh group respectively at 4 hours after blood transfusion (P〈0.01).Conclusion Blood transfusion promotes the expressions of TH1 and TH2 cytokines.Compared wi
