分化抑制因子(Id)又称DNA结合抑制因子,是螺旋-环-螺旋(helix-loop-helix,HLH)蛋白家族中参与负性调节的转录因子,具有抑制细胞分化、促进细胞增殖的作用。众多实验表明,Id蛋白在肿瘤中高表达并与肿瘤侵袭转移等恶性潜能密切相关,提示Id蛋白有助于判断肿瘤的预后。恶性肿瘤中Id蛋白的表达异常,可能是肿瘤侵袭性表型、预后差的标志。而通过靶向治疗抑制Id蛋白的表达可以有效抑制肿瘤增殖、侵袭转移。本文就近年来Id蛋白对肿瘤预后的相关因素,如恶性增殖、侵袭转移、血管生成等方面的影响机制及临床领域的研究进展作一综述。
Id proteins,or inhibitors of differentiation/DNA binding,are negative regulators of helix-loop-helix( HLH) type transcription factor,and they can inhibit cell differentiation and promote cell proliferation. Many experiments have proven that high expression of Id proteins in tumors was significantly related to malignant behaviors of tumors,such as tumor invasion and metastasis,suggesting Id proteins can be used to predict the prognosis of tumors. Abnormal expression of Id protein in malignant tumor might be associated with the invasive phenotype and poor prognosis of tumors. Id protein-targeted therapy can effectively inhibit tumor proliferation,invasion and metastasis. This paper reviews the mechanism by which Id protein influences the prognostic factors,such as malignant proliferation,invasion and metastasis,angiogenesis and the researches on progress of related clinical researches.
