采用三维荧光光谱法,结合化学计量学中基于交替归一加权残差(ANWE)算法的二阶校正方法,实现了人血浆样品中川芎嗪和替米沙坦的同时快速定量分析。当组分数N取4时,ANWE算法解析获得的川芎嗪和替米沙坦的平均回收率分别为99.3±3.5%和96.8±2.0%。本文同时给出了此方法的检测限(LOD)与定量下限(LOQ),获得了满意的实验结果。此外,用上述方法对替米沙坦片中替米沙坦的含量进行了快速测定,所得结果与液相色谱方法结果进行比较,同样获得了令人满意的实验结果。本方法可以很好地解决目标分析物之间,以及目标分析物与基体内源物质之间因光谱严重重叠所引起的难分辨问题,用"数学分离"代替或简化"物理分离"及"化学分离",实验不需对样品进行复杂预处理,简便高效,可为临床医药分析提供新思路。
This paper proposed a novel approach for rapid quantitative analysis of ligustrazine and telmisartan in human plasma samples by the incorporation of excitation-emission matrix fluorescence and second-order calibration methodology based on the alternating normalization-weighted error(ANWE) algorithm.When the selected component number was set to 4,the average recoveries of ligustrazine and telmisartan were 99.3±3.5% and 96.8±2.0%,respectively.Moreover,the accuracy of the ANWE method was also evaluated through the figures of merit,such as the limit of detection(LOD) and the limit of quantitation(LOQ).Meanwhile,the method was also utilized to quantify telmisartan in tablets and the result was validated by the high-performance liquid chromatography with fluorescence detection.The experimental results demonstrated that the ANWE method,as promising quantitative alternative,can be satisfactorily applied to the complex samples,which fully exploited "second-order advantage".This suggested method,which avoiding tedious pretreatment and having simple and fast analytical characteristics with good results,may be expected to provide a new powerful tool for clinical medicine analysis.
