目的探讨二甲双胍对过氧化氢(H_2O_2)所致脑片损伤的保护作用。方法通过H_2O_2孵育制备小鼠脑片损伤模型,预先给予不同浓度的二甲双胍进行干预。观察小鼠脑片TTC染色情况,测定TTC染色产物formazan的含量及脑片LDH的释放率。结果 H_2O_2可导致脑片TTC染色变浅、formazan含量减少及LDH含量增加,预先给予二甲双胍可剂量依耐性的增加脑片染色、增加formazan含量及减少LDH含量。结论二甲双胍对H_2O_2所致的脑片损伤有一定的保护作用。
Objective To observe the protective effect of metformin on brain slice injury induced by hydrogen peroxide in mice. Methods The brain slice injury model was induced by incubation with hydrogen peroxide and was treated with different concentrations of metformin. TTC staining,the contents of formazan and LDH were tested to assess the brain slice injury. Results Hydrogen peroxidereduced the color of TTC staining and the content of formazan,increased the content of LDH. Administration of metformin dose-dependently increased the color of TTC staining and the content of formazan,decreased the content of LDH. Conclusion Metformin has protective effect on brain slice injury induced by hydrogen peroxide.
