目的:鉴定LRP16与ART-27的相互作用,推测LRP16基因在肿瘤发生、发展中涉及到的信号途径。方法:酵母双杂交系统筛选出ART-27可与LRP16相互作用,采用酵母双杂交回复验证、免疫共沉淀及GST pull-down等体内、体外方法对二者之间的相互作用进行验证。结果:LRP16与ART-27在体内、体外均可形成复合物。结论:ART-27是LRP16的特异性结合蛋白,二者的相互作用为进一步研究LRP16在胚胎发育及肿瘤发生、发展中的功能及信号传导途径奠定了基础。
Objective:To identify the interaction between human LRP16 and ART-27,and to speculate the signal pathway of LRP16 on the development and progression of cancer.Methods: The interaction of ART-27 with LRP16 was found by yeast two-hybrid screening.And this interaction was tested by co-immunoprecipitation analysis in vivo and GST pull-down assay in vitro.Results: LRP16 could bind ART-27 in vivo and in vitro.Conclusion: ART-27 might be a binding partner of LRP16.The interaction between these two proteins may establish a foundation for further studying the function of LRP16 on embryonic development and cancer.
