目的:研究TRIM22蛋白对A型流感病毒复制能力的影响。方法:RT-PCR扩增人TRIM22全长编码序列并制备TRIM22真核表达质粒;通过绘制病毒生长曲线及直接做空斑的方法检测外源性表达TRIM22情况下,两种不同流感病毒株PR8(H1N1)和A/ST/364/2005(H3N2)在MDCK细胞中的复制能力。结果:成功构建TRIM22真核表达质粒;两种不同亚型流感病毒在过表达TRIM22的情况下,其在MDCK细胞中的生长曲线及空斑大小与阴性对照组相似。结论:TRIM22不能有效抑制不同亚型A型流感病毒的复制。
Objective:To investigate the effect of TRIM22 protein on the replication of influenza A viruses.Methods:Full-length encoding sequence of human TRIM22 was cloned by RT-PCR and a recombinant plasmid expressing TRIM22 was generated.Two different influenza A viruses(PR8(H1N1)and A/ST/364/2005(H3N2))were used for the subsequent analysis of viral growth curves and plaque sizes in MDCK cells upon the exogenous expression of TRIM22 or not.Results:A eukaryotic plasmid expressing human TRIM22 protein was successfully constructed.Influenza A viruses exhibited the similar growth kinetics and plaque phenotypes in MDCK cells with or without the overexpressed TRIM22.Conclusion:TRIM22 has no obvious inhibitory effect on the replication of different influenza A viruses.
