目的:检测Yes-associated protein ( YAP)相关蛋白及lats基因在人结肠癌组织中的表达,探讨其与结肠肿瘤发生发展的相互关系及临床意义。方法:应用HE染色和免疫组织化学方法检测32例结肠瘤样组织、52例结肠癌组织中YAP相关蛋白及其相关基因如lats基因的表达情况。结果:免疫组织化学方法检测结果显示YAP基因及其相关基因如lats基因主要是在结肠腺上皮组织中的细胞核中的表达呈阳性。 YAP及lats基因在结肠癌样组织中的表达与瘤样组织发生发展分级程度相关。结论:YAP及lats基因参与结肠瘤样组织的发生发展过程,并且有望为结肠瘤样组织的早期诊断,预防,临床治疗和预后判断提供新的靶点和可靠依据。
Objective:To detect the expression of Yes -associated protein ( YAP) and large tumor suppressor gene ( LATS) in hu-man colon cancer and to explore the relationship between them and the development of colon cancer and it ’ s clinical significance . Methods:HE dyed and immunohistochemical method are applied to detection of yap genes and their related genes such as lats gene ex -pression in 32 cases of colorectal adenoma and 52 cases of colorectal adenocarinoma .Results:the immunohistochemical method to de-tect the result shows that YAP genes and their related genes such as lats are mainly in the nucleus of coloretal epithelial tissues expres -sion was positive .Twenty cases of normal colon tissue , 26 cases of adjacent non -tumor tissues , 5 cases of colorectal polyps , 5 cases of adenoma of colon and 30 cases of colon cancer are positive .Comparison the five groups , the difference is statistically significant ( p〈0, 05).YAP and lats gene expression in colon cancer tissues are related to the development of tumor classification while there is no statistical difference in the clinical factors such as age , gender , and histologic grading .Conclusion:YAP and Lats genes are involved in colon cancer development process of the organization .They are expected to be reliable basis for the early diagnosis of colon cancer and provide new target for the early diagnosis , prevention , clinical treatment and prognosis judgment .
