目的体外培养、分选出胶质瘤干细胞,观察其生长方式并检测其细胞周期。方法体外原代无血清培养(添加EGF、FGF、B27等因子)人胶质瘤细胞和U87细胞株,观察其增殖分裂及多向分化,CD133流式分选后用碘化丙啶法检测其细胞周期。结果细胞出现2种分裂方式:对称分裂、不对称分裂。对称分裂后细胞一般不再增殖,而不对称分裂者逐渐形成细胞球。分选后细胞周期检测发现CD133+细胞qbGo/G,期比例高而CDl33-中G2/M期比例高(尸〈0.01)。结论不对称分裂是干细胞主要增殖方式,而肿瘤干细胞可分为静止群和活跃群,且静止群较多,可能是胶质瘤难以根治和易复发的原因之一。
Aim To analyze the cell cycle of CD133+ tumor stem-like cells (TSCs) from human malignant glioma. Methods Three human malignant glioma cell lines, SHG62, SHG66, and U87, were cultured in DMEM/F 12 medium containing EGF and bFGF. Proliferation and multilineage differentiation of these cells were investigated by limited dilution analysis and immunocytochemistry. CD133~ cells were collected by fluorescence activated cell sorting. The cell cycle of CD133~ cells was analyzed by flow cytometry using propidium iodide staining. Results SHG62, SHG66, and U87 cells were able to propagate, differentiate to multiple lineages. Both symmetric and asymmetric cell divisions were observed. CD133~ tumor stem-like cells were successfully separated from SHG62, SHG66, and U87 cell lines. The cells in Go/ G1 phase among the CD133~ cell group were significantly higher than those in the CD133- cell group, while cells were more distributed in G2/M phase among the CD133 group. Conclusion SHG62 and SHG66 cells display characteristics of tumor stem-like cell. Asymmetric division is the primary mode of proliferation. CD133+ cells tend to stay in G0/GI phase, indicating their quiescent nature.
