目的比较IL-4树突状细胞(DC)及IFN-α DC负载自噬小体疫苗后激活人抗原特异性T细胞的活性。方法用人单核细胞体外培养IL-4DC及IFN-α DC;制备表达CEF蛋白的HEK293T细胞,从中提取自噬小体疫苗DRibbles。分别用两种体外培养的DC负载HEK293TCEFDRibbles后刺激自体T淋巴细胞。用流式细胞术细胞内染色方法检测分泌IFN-γ的T细胞占总T细胞的百分比;流式细胞术表面染色方法检测两种DC表型差异。结果负载HEK293T CEF DRibbles的1FN-α DC能够诱导(6.0±1.2)%的自体CD8+T细胞激活,高于IL-4DC诱导的(2.3±0.6)%(P〈0.05),CD4+T细胞有效激活率亦高于II,4DCE(2.6±0.5)%VS.(0.3±0.1)%](P〈0.05)。与IL-4 DC相比,IFN-αDC细胞体积小,颗粒复杂程度低,表达较低水平的CD14、CD11c,但HLA-DR的表达水平类似。结论与IL-4 DC相比,IFN-α DC负载DRibbles能更有效激活人抗原特异性CD8+ T及CD4+T细胞。
Objective To compare the activation of human antigen-specific T cells stimulated by two different ex-vivo cultured DCs loaded with autophagosome vaccine(DRibbles). Methods The IL-4 DC and IFN-α DC were cultured from human monocytes in vitro. The DRibbles were extracted from HEK293T cells expressing CEF protein. Then the two different DCs loaded with HEK293T CEF DRibbles were used to stimulate autologous T cells. The percentages of T cells secreting IFN-γ were detected by flow cytometry intracellular staining. The phenotypes of two DCs were detected by flow cytometry. Results The percentage of autologous CD8+ T cells stimulated by IFN-γ DC loaded with HEK293T CEF DRibbles was higher than that of by IL-4 DC [(6.0± 1.2)% vs. (2.3± 0. 6)%] (P〈0.05). So did the effective activation rate of CD4+ T cells [(2.6±0.5)% vs. (0. 3±0.1)%] (P〈0. 05). Compared with the IL-4 DC, the IFN-γ DC was smaller in size and lower in complexity, but similar in expressing the lower levels of the CD14 CDllc and HLA-DR. Conclusion IFN-α DC loaded with DRibbles can activate human antigen-specific CD8+ T and CD4+ T more efficiently than IL-4 DC.
