目的研究七叶皂苷A的大鼠血浆蛋白结合率。方法在体外用平衡透析法模拟七叶皂苷A体内与血浆蛋白结合的过程,用高效液相色谱-质谱联用(HPLC-MS/MS)法测定低、中、高3个浓度的七叶皂苷A及其转化产物七叶皂苷C在透析袋内血浆中和透析袋外缓冲液中的浓度,计算七叶皂苷A的血浆蛋白结合率。结果低、中、高3个浓度的七叶皂苷A在体外与大鼠的平均血浆蛋白结合率分别为95.8%,96.6%和95.9%,3个浓度之间的血浆蛋白结合率比较差异无统计学意义(P〉0.05)。结论七叶皂苷A与大鼠血浆蛋白高度结合。
Objective To study the plasma protein binding rate of escin A in rats'plasma. Methods Under three different concentrations, this paper used equilibrium dialysis method to imitate the binding process be- tween escin A and the plasma protein, and determined the concentration of escin A and escin C (a transformation product of escin A) in and out of the dialysis membrane using high performance liquid chromatography - tandem mass spectrometry (LC -MS/MS) method and calculated the plasma protein binding rate of escin A. Results The plasma protein binding rate of escin A in rats was 95.8% , 96. 6% and 95.9% at low, middle and high concentration, respectively. There was no significant difference among three concentrations. Conclusion Escin A was highly bound to plasma proteins in rats.
