目的:探讨5-氮杂-2-脱氧胞苷(5-Aza-CdR)干预对胰腺癌细胞株PANC-1增殖和凋亡的影响及可能的机制。方法:用不同浓度的5-Aza-CdR处理PANC-1细胞株,未予干预的细胞为对照,采用台盼蓝染色、细胞计数法检测5-Aza-CdR干预前后细胞增殖能力的变化,FCM法检测干预前后细胞凋亡率的变化,caspase-3活性测定检测5-Aza-CdR诱导凋亡的能力。结果:5-Aza-CdR干预后,PANC-1细胞增殖明显受到抑制,细胞凋亡率明显提高,细胞内caspase-3活性明显增强(P〈0.05)。结论:5-Aza-CdR可以明显抑制胰腺癌细胞株PANC-1的生长增殖,并诱导细胞发生凋亡,线粒体凋亡途径可能是其诱导凋亡的机制之一。
Objective:To investigate the effects of 5-Aza-2-deoxycyt(5-Aza-CdR) on the proliferation and apoptosis of pancreatic carcinoma cell line Panc-1,and explore its possible mechanisms.Methods:PANC-1 cell line was treated by 5-aza-CdR of different concentrations.Trapan blue exclusion cell count assay,flow cytometry and activity test of caspase-3 protein were used to determine the cell growth and apoptosis.Results:After treated by 5-Aza-2-deoxycyt,the cell proliferation of PANC-1 cell line was inhibited obviously,the apoptosis rate was increased significantly(P0.05),compared with the control group.The activity of caspase-3 protein in cells was markedly enhanced after being treated by 5-Aza-2-deoxycyt.Conclusion:5-Aza-2-deoxycyt can inhibit cell proliferation of PANC-1 cell line,and induce apoptosis of PANC-1 cell line,the mitochondrial pathway might be one of mechanisms inducing the apoptosis of PANC-1 cells.
