左归丸和右归丸对冷冻大鼠卵巢组织存活效果的影响

目的评价左归丸和右归丸对大鼠卵巢组织冷冻效果的影响。方法在大鼠卵巢组织冷冻保存过程中,在冷冻保护剂中分别加入5%、10%、15%左归丸和右归丸含药血清和空白血清,比较各组解冻复苏后卵巢组织结构特点、卵泡正常形态率、卵母细胞凋亡率。结果新鲜卵巢组的卵泡正常形态率高于各冷冻卵巢组,卵母细胞凋亡率显著低于所有冷冻组(P〈0.05);10%、15%左归丸和右归丸含药血清组的卵泡正常形态率高于5%左归丸和右归丸含药血清组和空白对照组,卵母细胞凋亡率低于5%左归丸和右归丸含药血清组和空白对照组(P〈0.05)。结论左归丸和右归丸能提高冷冻卵巢组织的卵泡成活率,降低冷冻卵巢组织卵母细胞的凋亡率,为补肾中药成功应用于卵巢组织冷冻保存中提供了实验依据。

Objective To evaluate the effects of Zuogui pill (ZGP) and Yougui pill (YGP) on cryopreservation of rat ovarian tissues. Methods Female rats were divided into eight groups: 5%, 10%, 15%ZGP cryopreservation groups and 5%, 10%, 15% YGP eryopreservation groups, vacuity control group and fresh ovarian group. The percentages of normal follicle and the oocyte apoptotic rate were compared among all groups. Results The normal morphological follicles rate of all eryopreservation groups were lower than fresh ovarian group (P〈0. 05). The normal morphological follicles rate of cryopreserved ovarian tissue in 10%, 15% ZGP and YGP groups were higher than 5% ZGP, YGP groups and vacuity control group. The oocyte apoptotic rate of the fresh ovarian tissue were significantly lower than all cryopreserved groups ( P 〈 0. 05 ). The ooeyte apoptotic rate of eryopreserved ovarian tissue in 10%, 15% ZGP and YGP groups were significantly lower than 5% ZGP, YGP group and vacuity control group(P 〈 0. 05). Conclusion ZGP and YGP significantly improve the follicle survival rate of cryopreserved rat ovarian tissues. This study will establish the foundation for the application of traditional chinese kidney tonify herbs in ovarian tissue cryopreservation.
