目的:观察疏肝、健脾、活血、祛湿、综合不同治法方药对脂肪肝大鼠超微结构及Kupffer细胞CDl4mRNA表达的影响。方法:利用高脂饮食、白酒灌胃复制大鼠脂肪肝实验动物模型,同时给予疏肝、健脾、活血、祛湿、综合不同方药进行干预,12周后观察各组大鼠超微结构的改变,同时以Ⅳ型胶原酶、蛋白酶E联合灌注消化、梯度离心、选择性贴壁分离不同组别大鼠Kupffer细胞,采用半定量RT-PCR方法检测不同组别大鼠Kupffer细胞CD14mRNA的表达。结果:各组中药治法对NAFLD细胞超微结构的改变均有不同程度的修复作用,其中尤以健脾组明显。模型组大鼠Kupffer细胞CD14mRNA蛋白的表达较正常组明显增加(P〈0.05),各干预组CDl4mRNA表达较模型组有所降低(P〈0.05),其中健脾组最为明显。结论:模型组大鼠超微结构改变与NAFLD超微结构相吻合,NAFLD模型大鼠Kupffer细胞CD14mRNA表达明显增高,表明Kupffer细胞CD14mRNA表达明显增高可能为脂肪肝发病原因之一,抑制Kupffer细胞CD14mRNA表达可能是健脾等治法方药抗实验性大鼠脂肪肝的作用机制之一。
OBJECTIVETo investigate the effects of different therapeutic methods and typical recipes on ultra-structure of fatty liver and the activation of CD14 mRNA in Kupffer cells of rats. METHODS The rat model of fatty liver was established by feeding high fat diet combined with distillate spirit. Meanwhile Chinese medicines Shugan fang, Jianpi fang, Huoxue fang, Qushi fang, and Zonghe fang were given to treat different groups respectively. 12 weeks later, the Kupffer cells were isolated from livers of control group, model group and different treatment groups by sequential in situ perfusion with collagenaseIgand pronase E, density gradient centrifugation, selective adherence. The expressions of CDI4 mRNA in Kupffer cells of control group, model group and different treatment groups were detected by RT-PCR. RFSULTS The rat fatty liver experimental ani- mal models were successfully copied. In NAFLD rats of the model group, ultra-structural pathological features of the lipid ag- gregation showed the decrease of perinuclear lysosomal, the swelling of mitochondria and disorders of endoplasmic retieulum. Chinese medicine had different degree of repairing effects on ultra-structural pathological changes, especially in Jianpi group. The expression of CD14 mRNA protein in Kupffer cells of model group was obviously higher than that of the control group(P 〈0. 05). The expression of CD14 mRNA was lower in treatment group than that of the model group(P〈0. 05), especially in Jianpi group. CKINCLUSION The rat model of fatty liver was established and proved to be successful. The ultra-structure of fatty liver of rat model changed obviously. The high expression of CDI4 mRNA may be involved in fatty liver and contributed to liver injure. The recipes of Jianpi fang, Shugan fang, Qushi fang, Huoxue fang, and Zonghe fang may protect against liver injury by reducing CD14 mRNA expression in rats with fatty liver.
