为了解猪瘟病毒(CSFV)衣壳(C)蛋白在ST细胞中的定位情况,本研究将CSFVC蛋白基因克隆于真核表达载体pEGFP—C2中,构建重组质粒pEGFP—CSFV—C。将该重组质粒转染ST细胞,转染2h、4h和6h后利用激光共聚焦显微镜分析c表达蛋白在细胞中的定位情况。结果表明,CSFV表达蛋白在ST细胞中主要定位于细胞质和核仁。本研究为进一步分析CSFV的复制机理提供依据。
To study the localization of classical swine fever virus (CSFV) capsid protein in the swine testicle cells, the cells were transfected with recombinant plasmids of pEGFP-CSFV-C expressing capsid protein and examined using confocal microscopy at 2, 4 and 6 hours post transfection, respectively. The results showed that the CSFV capsid protein localized both in nucleolus and cytoplasm in the transfected cells, suggesting CSFV capsid protein had the ability of localization in nucleolus, which provided the basis for further studies of CSFV virus replication mechanism.
