噬菌体疗法在对杭多重耐药病原菌引起的感染方面具有很大的应用潜力。以耐万古霉素的屎肠球菌4P-SA为宿主菌分离筛选到一株烈性噬菌体,命名为m5。该噬菌体对不同的温度和pH值表现出稳定性,氯仿和乙醚不影响m5的感染能力。根据电镜观察到的形态特征表明m5属于有尾病毒目、长尾病毒科。噬菌体m5在体外对4P-SA具有强裂解作用;在体内,腹腔注射噬菌体m5 2.0×10^7 PFU即可有效保护菌血症状态的小鼠模型。本研究为利用噬菌体治疗多重耐药屎肠球菌感染奠定了基础。
Bacteriophage therapy could be an alternative to conventional antibiotic therapy against pathogens which are resistant to multiple antibiotics.In this study,a novel lytic phage was isolated from sewage,named m5,which can replicate and lyse highly vancomycin-resistant Enterococcus faecium 4P-SA.The phage has a certain stability against temperature and pH,and has a tolerance to chloroform and ethyl ether.The morphology characteristics of m5 suggest that it is a member of the Caudovirales,Siphoviridae family.Basic characterization demonstrated that the phage m5 lysed 4P-SA in vitro.A single injection of 2.0×10^7 PFU of the phage m5 was sufficient to protect the bacteremic mice.This finding provides more forcible evidence towards the therapeutic use of phages to combat multi-drug resistant E.faeciumbaeteria.
