以番茄果实为材料,比较了3种在果实中提取RNA的方法,针对番茄果实多糖的特点,利用无水乙醇和异丙醇联合沉淀的方法除去多糖,改良了Trizol法。为了检验含量比较低的miRNA,利用Licl沉淀的方法进行富集。结果表明:改良Trizol法能够有效地去除多糖,提取到的RNA 28S rRNA亮度约为18S rRNA的2倍,A260与A280比值为1.84,A260与A230比值为1.92,RNA产率为350.4μg/g,说明利用改良Trizol法从番茄果实中提取的RNA质量好、完整性好。由此法所得的RNA可直接用于cDNA文库构建等分子生物学实验。
Trizol method,guanidine thiocyanate method and perchlorate method,were employed to extract total RNA in fruit of tomato.And Trizol method was accordingly modified in extracting the total RNA in tomato by integrating a polysaccharide-removing step in correspondence with the fact that tomato fruit was rich in polysaccharides and polyphenol.Due to the low content of miRNA,we were using Licl precipitation method to enrichment it.The results showed that the modified Trizol method was capable of efficiently removing polysaccharides,and that of the RNA extracted by the modified Trizol method,the lightness of 28 S RNA was two times as much as that of 18 S RNA,the value of A260/A280,A260/A230 was1.84,1.92;and the RNA yield reached 350.4μg/g,which explained that the modified Trizol method could extract high-quality and high-complete RNA at a higher productivity.This high quality RNA could meet the demands of molecular analysis,such as cDNA library construction.
