目的:研究基因中抑制序列对离子通道基因表达的影响.方法:我们将hKv4.3基因的启动子和5’非翻译区的一段序列(+2-+160,S160)以不同的位置和方向克隆到报告质粒pβ-gal-Basievector中,瞬时表达,进行β—galactosidase相对活性分析.结果:S160对hKv4.3基因表达具有强烈的抑制作用.结论:抑制序列S160对hKv4.3基因的抑制功能具有位置特异性.
Objective:To study the effect of repressor on ion channel gene expression. Methods:We cloned hKv4.3 promoter and the sequence( + 2 - + 160, S160)of 5'UTR of hKv4.3 gene into pβ-gal-Basic vector. Transient expression of the pβ-gal vector and analysis of relative activity of β-galactosidase were undergone. Results: S160 intensively repressed the expression of hKv4.3 gene. Conclusion: We found that the inhibition of S160 to hKv4.3 gene with positionspecificity.
