目的探讨二甲双胍对甲状腺乳头状癌IHH-4细胞增殖的影响及其机制。方法 MTT法分别检测不同浓度的二甲双胍(0、5、10mmol/L)和二甲双胍10mmol/L在不同时间点(0、24、48、72h)对乳头状癌IHH-4细胞增殖的影响。Western blot法检测不同浓度二甲双胍干预IHH-4细胞48h后单磷酸腺苷激活的蛋白激酶α亚基(AMPKα)及磷酸化AMPKα(p-AMPKα)蛋白表达。结果二甲双胍可呈浓度和时间依赖性抑制IHH-4细胞增殖(P〈0.01)。随着二甲双胍干预浓度的增加,p-AMPKα/AMPKα表达量增加(P〈0.01)。结论二甲双胍可通过激活AMPK通路抑制甲状腺乳头状癌IHH-4细胞的增殖活性。
Objective To explore the effect and mechanism of metformin on the proliferation of papillary thyroid carcinoma IHH-4 cells. Methods MTT assay was used to measure the proliferation of IHH-4 cells treated by different concentrations of metformin 0, 5 and 10 mmol/L and metformin 10 mmol/L at different time points of 0,24,48 and 72 hours. Western bolt was performed to detect the expressions of AMPKa and phospborylated AMPKa(p-AMPKa) of IHH-4 cells treated by different concentrations of meformin at 48 hours. Results Metformin could significantly inhibit IHH-4 cell proliferation in the concentration- and time-dependent manners(P〈0. 01). With increasing concentrations of metformin, the expression level of p-AMPKa/AMPKa was significantly increased (P〈0. 01 ). Conclusion Metformin can inhibit the proliferation activity of papillary thyroid cancer IHH-4 cells via activating AMPK pathway.
