研究目的是探讨电穿孔技术在意大利蜜蜂卵基因转化中应用的可能性。实验用质粒pEGFP-Nl对意大利蜜蜂卵电转导条件优化进行系统研究,然后利用所得到的电穿孔优化条件,对大量意大利蜜蜂早、晚期卵进行绿色荧光蛋白基因转移研究。实验结果获得1组较佳电学参数:脉冲电压750 V/cm,脉冲时间160μs,电击次数1;意蜂早期卵阳性表达率为0.31%~0.90%,晚期卵为0.00%~0.30%。结果表明电穿孔法可以促使意大利蜜蜂卵摄入外源基因,且意大利蜜蜂早期卵电转导效果比晚期卵好。
The object of the study was to explore the possibility of the application of electroporation in gene transformation of the Apis mellifera ligustica eggs.A systematic study on the transformation conditions for the eggs was conducted with the plasmids of pEGFP-N1 by using electroporation.Great quantities of early or late Apis mellifera ligustica eggs were electroporated with green fluorescent protein gene on the optimized conditions.A group of highly optimized parameters such as electrical voltage(750 V/cm),pulse time(160 μs) and number of pulses(1) were abtained.The exogenous gene expression rates for early and late Apis mellifera ligustica eggs were 0.31%~0.90% and 0.00%~0.30% respectively.These findings suggested that electroporation could force the foreign gene to enter Apis mellifera ligustica eggs,and the transformation efficiency for the early eggs was higher than that of the late eggs.
