肿瘤耐药是当前肿瘤治疗的主要障碍。近年来,包括药物外排增强、药物亚细胞水平分布改变、药物亲和力改变、细胞解毒和修复功能加强及肿瘤细胞抗凋亡能力增强等肿瘤耐药相关生物学机制的研究已取得很大进展,这些生物学机制共同构建成一个复杂的网络系统,详尽阐明这一多因子的网络能够更好地预测肿瘤患者对治疗的应答及制定肿瘤患者的个体化治疗。应用蛋白质组学技术已在肿瘤细胞系和组织中鉴定出大量新的耐药相关性因子和蛋白表达标记物。本文就比较蛋白质组学研究方法在不同肿瘤细胞中所发现的耐药相关蛋白及机制进行综述与分析。
Drug resistance is a major obstacle for cancer chemotherapy.In recent years,various biological mechanisms associated with drug resistance have been identified including increased drug efflux or detoxification,enhanced DNA repair,decreased drug binding,increased anti-apoptotic factors,etc.To elucidate this multifactorial network is helpful for predicting therapeutic response and prescribing individual-tailored therapeutic regime.Various new factors or proteins involved in drugresistance have been identified from cancer cell lines and cancerous tissues by using proteomic technique.In this article we will focus on studies of proteomics in anticancer drug resistance and discuss some common mechanisms.
