目的明确白及非多糖部分的止血作用,初步了解其止血机制.方法白及95%乙醇提取物上D101大孔树脂,经水洗脱后,用80%乙醇洗脱得到不含多糖的80%乙醇组分(BS-80EE);以小鼠全身肝素化的出血时间(BT)和凝血时间(CT)为指标考察BS-80EE的止血作用;以二磷酸腺苷(ADP)为诱导剂,采用比浊法观察BS-80EE对大鼠体内血小板聚集率的影响;通过考察BS-80EE对大鼠凝血酶时间(TT)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)和纤维蛋白原含量(FIB),以及血小板膜糖蛋白P-选择素(P-S)、凝血酶-抗凝血酶复合物(TAT)、纤溶酶原激活物抑制剂-1(PAI-1)、D-二聚体(D-D)的影响,分别从血小板和凝血纤溶系统角度了解BS-80EE的止血机制.结果BS-80EE可剂量依赖性地缩短全身肝素化小鼠的CT和BT(P〈0.01或P〈0.05);明显促进ADP诱导的大鼠体内血小板聚集作用(P〈0.01);明显缩短大鼠的TT值(P〈0.01或P〈0.05);高剂量组可明显增加FIB含量(P〈0.05);中、高剂量组可明显增加P-S、TAT、PAI-1的含量,减少D-D的生成(P〈0.01).BS-80EE虽有缩短大鼠APTT和PT的趋势,但差异均无显著性.结论白及非多糖组分(BS-80EE)具有良好的止血功效,可通过促进血小板聚集和凝血而发挥止血作用.
Aim To understand the hemostatic effect ofa non-polysaccharide fraction of Bletilh striatt ( BS -80EE ) and to clarify its mechanism of action. MetliodsThe non-polysaccharide fraction ( BS -80EE ) wasprepared by passing the 9 5 % ethanol extract of BletiHthrough a D 101 macroporous resin column elutedfirst with water and then with 8 0 % ethanol. Bleedingtime ( ADP ) and clotting time ( CT ) of heparinizedmice were employed as indicators for evaluating the hemostaticeffect of BS -80EE . The mechanism of actionwas investigated through observing the effect of B S -80E E on platelet aggregation induced by adenosinediphosphate ( ADP ) in rats with nephelometry and testingthe effect of B S -80E E on the thrombin time(TT ) ,prothrombin time ( PT ) ,activated partial thromboplastintime(APTT ) , fibrinogen ( FIB) ,P -selectin( P-S ) ,thrombin-antithrombin complex ( TAT ) , D-dimer ( D-D ) and plasminogen activator inhibitor-1 ( PAI-1 ) .Results B S -80E E significantly shortened the CT andADP ( P 〈0. 01 or 0. 05) of heparin mice in a dose-dependentmanner; groups of all doses significantly reducedthe rat TT ( P 〈 0. 01 or 0. 05 ), and the highdosegroupsignificantly increased theFIB content ( P 〈0.05); the mid-dose group and high-dose groups ofB S -80E E significantly increased the contents of P-S ,TAT and PAI-1,while reduced the D-D production inrats ( P 〈 0. 01 ) ; although dose-dependent reductionsof APTT and PT were observed for each treatmentgroup,no significance was observed. Conclusion B S -80E E possess pronounced hemostatic effect by promotingplatelet aggregation and coagulation.
