为发掘低分子量谷蛋白亚基(LMW-GS)新的变异类型,通过设计引物和特异PCR扩增,从簇毛麦TA2127基因组DNA中得到14个LMW-GS基因,GenBank登录号分别为HQ615147~HQ615160。序列分析表明,所有基因具有完整编码区,长度为831bp和846bp,可编码277和282个氨基酸残基,推导的氨基酸序列分析显示,14个基因编码的蛋白序列都缺少一段N-端保守区,且N-端序列是一种新的类型;在14个LMW-GS基因中检测到19个SNPs和1个16bp的缺失。系统进化分析表明,簇毛麦LMW-GS与ω-醇溶蛋白和HMW-GS亲缘关系相对较远,与普通小麦Glu3-的低分子谷蛋白基因进化关系相对较近。
In order to discover new LMW-GS genes and provide more important information for the further genetic improvement,fourteen novel LMW-GS genes(GenBank accession number: HQ615147~HQ615160) from Dasypyrum villosa(TA2127) were amplified and cloned using PCR-based strategy.The lengths of the open reading frames(ORF) of those LMW-GS genes were 831 and 846 bp,encoding a mature protein of 277 and 282 amino acid residues.Comparative analysis showed that all genes lacked the conserved sequence of N-terminal,and the N-terminal was new types.A total of nineteen single nucleotide polymorphisms(SNPs) and a 16 bp deletion among the fourteen LMW-GS genes were identified.Phylogenetic and evolutionary analysis revealed that the LMW-GS genes from Dasypyrum villosa were closely related to LMW-GS genes of Glu-3 and less homology was found between LMW-DV genes with HMW-GS and ω-gliadin genes.
