目的研究不同粒径纳米银颗粒对其杀灭铜绿假单胞菌的效果差异,为实现铜绿假单胞菌的杀灭奠定基础。方法合成不同粒径的纳米银颗粒,分别将这些纳米银颗粒与处于指数生长前期的细菌相混合,经过不同处理时间后取100μl加入LB培养基中培养18h后,运用CAM/EthD-1荧光染料进行细菌细胞死活分析。结果不同粒径大小的纳米银颗粒的杀菌效果不一致:颗粒越小,其杀菌和灭菌的效果越好,经10nm的纳米银颗粒作用5min可杀灭80.0%的铜绿假单胞菌。结论不同粒径纳米银颗粒及浓度对其杀灭铜绿假单胞菌的效果不同,颗粒越小,其杀菌和灭菌的效果越好,从而为临床微生物的杀灭提供了一种新的技术方案。
OBJECTIVE To investigate the effects of various diameters of silver nanoparticles on killing Pseudo- rnonas aeruginosa so as to provide bases for killing P. aeruginosa. METHODS The different diameters of silver nanoparticles were synthesized and introduced to kill P. aeruginosa. After incubation for a series of time, 100μl of mixture was taken out to inoculate for 18h to count the colony-forming unit. The CAM/EthD-1 stain was used to determine the killing effect of the silver nanoparticles. RESULTS The smaller the silver nanopartides were, the more powerful the killing effect was. The 10nm diameter silver nanoparticle could kill 80.0% of the P. aeruginosa in 5 min. CONCLUSION The different diameters or concentrations of silver nanoparticles have various effects on killing P. aeruginosa, the smaller the particle, the better the effect of the sterilization is, thus it has provided a new approach to kill the clinical microorganisms.
