目的制备阳离子表面活性剂型杀菌剂度米芬的微米纤维膜,考察不同载药量时度米芬的释放行为。方法采用均相溶液静电纺丝法制备载度米芬质量分数为1.25%、2.5%、5%的聚L-乳酸纤维,广角X-射线衍射法和差示热分析法考察度米芬在聚L-乳酸纤维表面及内部有无析出。将纤维膜置于水中释放度米芬,不同时间点取释放介质并采用高效液相色谱法测定释放介质中度米芬的含量,绘制时间-释放百分率曲线图。结果度米芬和聚L-乳酸混合溶解后能够静电纺成粗细较为均匀的纤维,纤维直径随载药量的升高而升高。纤维表面和内部无晶态度米芬析出。药物有明显的突释,在突释后至5 h呈现缓释行为,并随之进入平台期。相同时间点药物释放百分率随着载药量的增高而降低。去除突释点后,释放百分率-释放时间曲线对Freundlich方程有较高的拟合值。结论聚L-乳酸电纺纤维包裹度米芬后对后者呈现特别的释放行为,对类似结构小分子表面活性剂从电纺纤维的释放行为具有参考意义。
Objective To prepare micrometer-scale fibrous membrane containing cationic surfactant antibacteria agent domiphen.And to investigate the release behavior of domiphen of different loading amount.Methods Poly L-lactic acid fibers with 1.25%,2.5% and 5%(w) domiphen loaded were prepared by homogeneous solution electrospinning method.Wide angle X-ray diffraction and differential thermal analysis were used to investigate whether or not there was drug crystals separated on the fiber surface or within fibers.The fibrous membranes were immersed into water to release domiphen,and at different sampling time,the release medium was drown and quantified for domiphen amount in release medium by high performance liquid chromatography from which time-release percent curves were plotted.Results Mixture solution of domiphen and poly L-lactic acid could be electrospun into fibers of homogeneous diameters which diameters increased as the amount of domiphen increased.There was no drug crystal separated out on the fiber surface and within fibers.There was significant burst release at the beginning,5 h sustained release afterwards,and went into plateau till 24 h during the release process.Except the burst release point,release percent-time curve had rather high fitting value to Freundlich' equation.Conclusions Poly L-lactic acid fibers with domiphen entrapped exhibit special release behavior which has value for referencing of small molecular surfactants of similar structure released from electrospun fibers.
