胆道闭锁患者肝内胆管γδT细胞和调节性T细胞浸润及意义

目的 研究胆道闭锁(Biliary atresia,BA)患者肝组织中γδT细胞和调节性T细胞(Foxp3^+Treg)的比例变化。方法 采用免疫组织化学方法和流式细胞术观察和检测胆道闭锁患儿组(BA组)23例和对照组(CG组)12例肝组织中γδT细胞分布情况以及γδT细胞和Foxp3^+ Treg细胞比例关系。结果 免疫组织化学染色显示BA组肝脏汇管区胆管周围有大量γδT细胞和一定程度的Foxp3^+ Treg细胞浸润。流式细胞术显示胆道闭锁肝组织中γδT细胞与Foxp3^+Treg细胞比例明显高于对照组(P〈0.05),且γδT细胞与Foxp3^+Treg细胞比例呈显著负相关(P〈0.05)。结论 胆道闭锁患儿肝组织中γδT细胞增多,或抑制Foxp3^+Treg细胞增值,促进了胆管的进行性炎症损伤。

Objetive To investigate the changes of the percentages of γδT cells and Foxp3^+ Treg cells in the liver tissues of patients with biliary atresia(BA). Methods γδ ceils and Foxp3^+ Treg cells were determined in the portal tracts of patients of 23 cases BA patients and of the 12 cases from the control group using immunohistochemical staining and flow eytometry. The liver tissues lymphocytes were separated with Percoll Hypaque density gradient centrifugation. The proportion of γδT cells and Foxp3^+ Treg cells were detected with flow cytometry. Results γδT cells and Foxp3^+ Treg cells are scattered in the interface areas of inflamed portal tracts of BA livers. BA tissues contained more γδT cells and Foxp3^+ Treg cells. The percentage of γδT cells in the liver of BA patients were significantly elevated compared with the healthy controls. The percentage of Foxp3^+ Treg cells decreased in the liver of BA patients compared with healthy controls. There was significant negative correlation between the percent of γδT cells and the percent of Foxp3^+ Treg cells in BA patients ( P 〈 0.05). Conclusions The up-regulation of liver γδT ceils in BA patients might cause the down-regulation of liver Foxp3^+ Treg cells ,thus contributes to the occurrenee of the disease.
