【目的】研究白藜芦醇减轻高原缺氧小鼠认知功能障碍以及对小鼠高原脑水肿的保护作用。【方法】昆明种小鼠随机分为5组:正常对照组、急性高原缺氧组、白藜芦醇大、中、小剂量预处理组;采用不同浓度的白藜芦醇预处理小鼠1周,将其放入低压氧舱24 h,模拟海拔8 000 m、湿度、温度以及12 h白昼。应用干湿法测定其脑含水量,伊文思蓝含量法分析血脑屏障,Y型迷宫实验测定小鼠认知功能的改变。【结果】白藜芦醇预处理组小鼠脑含水量及伊文思蓝含量明显减少,中剂量时脑水含量及伊文思蓝含量最低;白藜芦醇预处理组小鼠认知功能明显改善。【结论】白藜芦醇可减轻高原缺氧对小鼠认知功能的影响,并对小鼠高原脑水肿具有保护作用。
【Objective】To study the effect of resveratrol on the alleviation of cognitive dysfunction in mice with high-altitude hypoxia and protection of high-altitude cerebral edema.【Methods】Kunming mice were randomly divided into 5 groups: normal control group,acute high-altitude hypoxia group, pretreatment group with reseratrol in high, medium or low dose. Mice were pretreated with different concentrations of resveratrol for a week, and placed in a hypobaric chamber for 24 h which mimicked the height, humidity and temperature of 8 000 m above sea level as well as 12:12 h day and night. Brain water content was detected with dry-wet method. Analysis of blood brain barrier was performed with Evans blue content method. Mouse cognitive function was determined with Y maze experiment.【Results】The brain water content and Evans blue level in mice significantly reduced in pretreatment group with resveratrol and were the lowest in the group with medium dose of resveratrol. Cognitive function in mice was significantly improved in pretreatment group with resveratrol.【Conclusion】Resveratrol can alleviate cognitive dysfunction in mice induced by high-altitude hypoxia and protect mouse high-altitude cerebral edema.
