为分离出含内源质粒的乳酸细菌,用平板划线法从民间腌制食品酸蒜苔中分离到多株菌落乳白色、表面光滑圆润的产酸菌株;接种所分离的产酸菌株至MRS液体培养基,收集菌体用SDS-碱裂解法抽提质粒,获得含内源质粒菌株P5371,经纸层析定性鉴定P5371为一株产乳酸细菌。常规鉴定确.定该菌株为革兰氏阳性、无芽孢、杆状、接触酶阴性、不运动的兼性厌氧乳杆菌,16S rDNA和API 50CH系统分析表明P5371属于戊糖乳杆菌(Lactobacillus pentosus)。
To obtain Lactic Acid Bacteria containing endogenous plasmid,some ivory and sleek acid-producing bacteria were isolated from pickled acid garlic by streak plate method.Plasmid-containing strain named P5371 was obtained by plasmid extraction analysis through SDS-alkaline lysis method from the acid-producing bacteria above.Paper chromatography analysis showed that the strain P5371 produced lactic acid conspicuously.For the strain P5371,a general test demonstrated that it is Gram-positive,no gemma,bacilliform,facultative anaerobic,without catalase and moving activity.16S rDNA analysis and API 50CH system test classify strain P5371 belonging to L.pentosus.
