植物COP9(Constitutively photomorphogenic 9)信号体(CSN)是一个多亚基复合体,而CSN8是这个复合体必不可少的亚基。为了克隆油菜CSN8基因并分析其在各种环境信号下的表达变化,以甘蓝型油菜中双9号为材料,采用同源克隆法克隆油菜CSN8(BnCOP9)cDNA。序列分析表明,BnCOP9编码的蛋白含有197个氨基酸残基,在其蛋白质中部含有1个PCI结构域,与Arabidopsis thaliana CSN8(AtCOP9)有92%的相似性。定量RT-PCR分析显示,Scle-rotinia sclerotiorum、伤处理以及甲基茉莉酸快速激活BnCOP9的表达,苯丙噻重氮抑制其表达,表明BnCOP9作为各种环境信号的响应基因在水杨酸和茉莉酸信号的交叉谈话中及油菜对S.sclerotiorum和机械伤害反应的网络中起作用,这些结果为研究BnCOP9在油菜抗逆中的作用提供线索。
Plant COP9(Constitutively photomorphogenic 9)signalosome(CSN)is a multi-subunit complex in which CSN8 is an obligatory subunit.To clone CSN8 and investigate its response to environmental cues in oilseed rape,a full length cDNA of Brassica napus CSN8(BnCOP9)was isolated from the line Zhongshuang9 by homology cloning approach.Sequence analysis showed that BnCOP9 contains a PCI domain in the central region and 197 amino acids exhibiting a 92% identity with Arabidopsis CSN8(AtCOP9).Quantitative RT-PCR analysis showed that BnCOP9 expression were promptly activated by Sclerotinia sclerotiorum,wound treatment and MeJA,while suppressed by Benzothiadiazole,suggesting that BnCOP9 is in response to environmental stresses and play a role in crossing-talk of SA-and JA-mediated signaling and the molecular network of oilseed rape response against both S.sclerotioru and wound.These results provide a cue to study a role of the CSN complex in defense responses in oilseed rape.
