目的观察左、右归丸及其拆方对去卵巢大鼠肾脏PPARγ、FABP4基因与蛋白表达的影响,并探讨补肾法对绝经后妇女脂代谢紊乱的改善。方法将90只SD雌性大鼠随机分为空白组、假手术组、模型组、补佳乐组、左归丸组、右归丸组、共同药组、滋阴药组、补阳药组。除假手术、空白组外均进行双侧卵巢切除。经12周治疗分别以Q-PCR法、Western blot法检测PPARγ、FABP4在肾脏中基因和蛋白的表达。结果与空白组相比,除假手术组外,PPARγ、FABP4 mRNA及蛋白表达升高(P〈0.01);与模型组比较,各用药组PPARγ、FABP4 mRNA及蛋白表达降低(P〈0.01);与补佳乐组比较,各用药组PPARγ、FABP4表达高,其中左归丸组表达较低。结论左、右归丸及其拆方可以明显影响肾脏中脂代谢相关指标,其中以左归丸改变最为显著。
Objective To observe Zuogui pill,Yougui pill and their disassembled prescriptions in ovariectomized rat kidney,and to explore to improve kidney postmenopausal women lipid metabolism disorder Methods Dividing 90 SD female rats to 9 groups randomly: Control group,Sham group,Ovariectomy group,BJL group,ZGW group,YGW group,GTY group,ZYYgroup,BYY group,10 rats in each group. Escape Control group and Sham group,other groups eccoped ovaries group. After 12 weeks. Use real- time quantitative PCR,Western blot to detecting the gene and protein expression about PPARγ、FABP4 in the kidney. Results Compared with Control group,the gene and protein expression of PPARγ,FABP4 is improved( P〈0. 01). Compared with OVX group,the gene and protein expression of PPARγ,FABP4 is lower( P〈0. 01),which ZGW group better. Conclusion Zuogui pill,Yougui pill and their disassembled prescriptions made PPARγ,FABP4 lower expression thus effective interventions ovariectomy- induced osteoporosis,which Zuoguipill more notable.
