目的本研究从γδT细胞识别的应激配体出发,探索肝癌细胞的分子标记物。方法流式细胞技术检测了3个人肝癌细胞系(HepG2、SMMC7721和Hep3B)和1个正常的人胚肝细胞系(ccc HEL-1)表面11种已知的γδT细胞识别的应激配体的表达情况。收集了9名肝癌患者和10个健康对照的外周血样品,流式细胞技术分析了患者和对照的γδT细胞及其亚型的比例,ELISA检测血浆中10种应激配体的质量浓度。结果 3种不同的肝癌细胞系应激配体的表达情况存在着差异,与正常胚肝细胞系相比,ULBP1、ULBP5、HSP60、HSP90和MSH2在3种肝癌细胞系表达都有上调;与正常对照相比,肝癌患者外周血的γδT细胞总比例显著升高,CD27~-γδT细胞升高,而外周血主要的Vδ2细胞比例显著降低;10种应激配体在血浆中的质量浓度差异很大,在血浆质量浓度较高的应激配体中,ULBP1、ULBP4和HSP90在肝癌患者血浆中较正常人升高,有统计学意义。结论γδT细胞识别的应激配体ULBP1和HSP90在肝癌细胞系表面上调表达,并且在肝癌患者血浆中的质量浓度显著升高,是肝癌比较可靠的分子标记物。
Hepatocellular carcinoma (HCC) is a common malignant tumor with a survival years. This study try to explore molecular markers of HCC, focusing on the stress-induced ligan cells. Flow cytometry was employed to detect the expression of 11 stress-induced ligands in rate of less than five d recognized by γδT the surface of three commonly used human HCC cell lines (HepG2, SMMC7721 and Hep3B) and a normal human embryonic liver cell line (cccHEL-1). Peripheral blood mononuclear cells (PBMC) and plasma were collected from 9 HCC patients and 10 healthy controls. The proportion ofγδT cells in PBMC was analyzed by flow cytometry and the concentration of 10 stress-induced ligands in plasmas were detected by ELISA. Data showed that there were differences in the expression pattern of stress-induced ligands among the three HCC cell lines. ULBP1, ULBPS, HSP60, HSP90 and MSH2 were up-regulated in all three HCC cell lines when compared with cccHEL-1. The total proportion of γδT cells and CD27-γδ T cells in PBMC of HCC patients were significantly increased, while the proportion of V82γδT cells was significantly decreased. The concentration of 10 stress-induced ligands in plasma were very different, and the concentration of ULBP1, ULBP4 and HSP90 in the plasma of HCC patients were significant higher than that of controls. Our results indicated that stress-induced ligands ULBP1 and HSP90, up-regulated in HCC cell lines and plasma of HCC patients, will be reliable molecular markers for HCC.
