目的:观察桃红四物汤对高同型半胱氨酸大鼠主动脉死亡受体途径凋亡的影响。方法:40只雄性SD大鼠随机分为正常组、模型组、桃红组、叶酸组,每组10只,正常组给予普通饲料,模型组、桃红组、叶酸组给予高蛋氨酸饮食(普通饲料添加2%蛋氨酸)喂养,桃红组采用桃红四物汤水煎液,按4.59 g生药/(kg.d)灌胃;叶酸组采用叶酸混悬液,按1.35 mg/(kg.d)灌胃;模型组及正常组给双蒸水2 mL/(只.d)灌胃。12周后,Elisa法检测血清hcy、TNF-α、MCP-1浓度,免疫组化检测主动脉Cleaved Caspase3表达,RT-PCR法检测主动脉TNF-α、Fas mRNA表达。免疫印迹法检测主动脉TNF-α的表达。结果:12周后,模型组大鼠血Hcy显著升高,血清TNF-α浓度升高,血清MCP-1浓度有升高趋势;主动脉内皮细胞连续性受损,受损部位下的血管平滑肌细胞Cleaved Caspase3呈片状黄褐色阳性染色。主动脉全层行RT-PCR示TNF-α、Fas mRNA表达上调,western blot示TNF-α表达上调。相对于模型组,桃红组和叶酸组大鼠血清TNF-α浓度降低,主动脉未发现片状Cleaved Caspase3阳性染色,TNF-αmRNA表达明显下调,Fas mRNA表达有下调趋势,TNF-α蛋白表达下调。但叶酸组伴有血同型半胱氨酸浓度的下降,而桃红组没有降低血同型半胱氨酸浓度的作用。结论:高同型半胱氨酸血症诱导SD大鼠主动脉死亡受体途径凋亡,桃红四物汤可降低血清TNF-α,并在基因和蛋白水平下调主动脉TNF-α表达。
Objective : To test the therapeutic effect of Taohong Siwu ( THSW ) decoction, a compound traditional Chinese herbal medicine: on Fas mRNA expression of hyperhomocysteinemia rats' aortae. Methods: Forty male SD rats were randomly divided into 4 groups with 10 in each group: untreated group, model group, THSW group and Folic Acid group. Thirty male SD rats received a high methionine diet to induce hyperhomocysteine and endothelial dysfunction. Rats in THSW group and Folic acid group were fed with THSW 4.59 g/(kg d), Folic Acid 1.35 mg/(kg ~ d) respectively. Rats in untreated group and model group were fed with distilled water 2 mL/d intragastrically. At the end of 12 weeks, serum homoeysteine levels were test by enzyme-linked immunosorbent assay ( ELISA ), plasma TNF- ot and MCP-1 levels were tested with commercial kits. Fas, TNF- ct mRNA were determined by RT-PCR. The ~xpressions of cleaved Caspase3 was assayed by Immunohistochemistry. The expression of TNF- cx of aortae were determined by western blot. Results : Compared with untreated group, Homocysteine in model group was obviously higher. The homocysteine levels in Folic Acid group were decreased obviously. Compared with untreated group, the levels of TNF- ot and MCP-1 in model group in serum were higher. The TNF- oL level in serum was lower than that in THSW goup significantly. In aorta, the mRNA levels of TNF- ot and Fas in model group were significantly heightened, THSW decoction can down-regulate the levels of aortic TNF- ot mRNA significantly. The expression of cleaved Caspase 3 and TNF- ot in model group were increased significantly compared with those in untreated group. However, the expression of cleaved Caspase 3 was downregulated in the THSW group. Conclusions : A high methionine diet can not only heighten serum homocysteinemia significantly but also induce the Fas mediated apoptosis. THSW decoction can down-regulate the expression of TNF-a mRNA in serum and reduce the aortic level of TNF- aat protein and gene level.
