目的:使用交流箱心理应激模型,研究心理应激对三叉神经节中降钙素基因相关肽(CGRP)表达的影响。方法:90只SD大鼠,随机分为3组:空白对照组、足部电击组和情绪应激组,采用反转录-聚合酶链作用(RT—PCR)技术,检测应激后不同时间点三叉神经节中CGRPmRNA水平的变化。结果:三叉神经节中CGRPmRNA的表达在应激第1天无显著性差异,后逐渐升高,第7天时达到最高峰.又逐渐降低,但在第14天是与正常对照组相比还是升高了(P〈0.05),去除应激后CGRP的表达逐渐降低,在恢复14d后基本达到正常水平。结论:心理应激可以引起三叉神经节CGRPmRNA表达发生变化,可能是心理应激导致咀嚼肌痛觉敏感的中枢致病因素之一。
Objective: To study the expression of CGRP mRNA in the rat trigeminal ganglion during emotional stress by using the equipment of communication box. Methods : 90 Sprague - Dawley rats were randomly divided into 3 groups : control group, emotional stress (ES) group and footshock (FS) group. FS group was only used for inducing emotional stress and was not concerned in the result of experiment. The expression of CGRP mRNA in trigeminal ganglion was detected by reverse transcription -polymerasa chain reaction (RT -PCR). Results: The CGRP mRNA expression of trigeminal ganglion in ES group was stronger than that in control group at most time points during emotional stress. CGRP mRNA level was increased after one day and reached the peak level on the 7th day. Although it started to gradually decrease after that, it still showed higher level than that of the controls till the 14th day after emotional stress. CGRP mRNA level was gradually decreased after removing the emotional stress, and back to normal in the 14th day after the removal of emotional stress. Conclusion : It is suggested that emotional stress can change the expression of CGRP mRNA in the rat trigeminal ganglion. This change may be one of the central mechanism of hyperalgesia of masticatory muscles under emotional stress.
