目的探讨厄贝沙坦对酒精性脑损伤大鼠血清及脑组织神经元特异性烯醇化酶(NSE)水平的影响。方法给予60%酒精4 mL灌胃建立酒精性脑损伤动物模型,将60只雄性SD大鼠,随机分为酒精组、对照组、药物组3组。酒精组和药物组给予60%酒精4 mL灌胃;药物组同时给予厄贝沙坦5 mg/d。对照组给予同等剂量的生理盐水。2周后抽取大鼠腹主动脉血液,断头取脑,采用酶联免疫法测定各组血清及脑组织中NSE水平。结果酒精组血清中NSE(7.322±1.406)μg/L和脑组织中(13.314±1.395)μg/L明显高于对照组(P〈0.05);药物组血清NSE(3.032±2.497)μg/L、脑组织中NSE(3.796±0.294)μg/L明显低于酒精组(P〈0.01)。结论厄贝沙坦可降低大鼠酒精性脑损伤中NSE水平,有望成为治疗酒精性脑损伤患者的预防用药。
Objective To explore the effect of irbesartan on the content changes of neuron specific enolase(NSE) in sera and brain tissues of rats with alcoholism brain damage.Methods Rat models of alcoholic brain damage were established by drinking 60% 4 ml alcohol.Sixty male SD rats were randomly divided into 3 groups: an alcohol group,a control group and an irbesartan group.The rats in the alcohol group and irbesartan group were given dehydrated alcohol(60%,4 ml),once a day,the rats in the irbesartan group were given dehydrated irbesartan(5 mg/d),and the rats in the control group were given normal saline once a day.After 2 weeks,all the rats were sacrificed and the NSE concentrations in sera and brain tissues were detected with ELISA.Results Compared with the control group,the contents of NSE [(7.322±1.406)μg/L and(13.314±1.395) μg/L] in the sera and brain tissues of the alcohol group obviously increased(P0.05).Compared with the alcohol group,the contents of NSE [(3.032±2.497) μg/L and(3.796±0.294) μg/L] in sera and brain tissues of the irbesartan group obviously decrease(P0.01).Conclusions Irbesartan can decrease the level of NSE in the rats with alcoholic brain damage.Therefore,it may be applied in the treatment of alcoholic brain damage.
