从长沙岳麓山和湘江等地富含油脂的样品中分离得到一株产脂肪酶菌株CS 1-1,经18S rDNA序列分析,确定该菌株为黑曲霉(Aspergillus niger).对该菌株的产酶条件进行优化,发现该菌株在以体积分数为1%的橄榄油为诱导物,5 g/L蔗糖为碳源,40 g/L蛋白胨为有机氮源,1 g/L(NH4)2SO4为无机氮源,pH为6.5的培养基中,于28℃,180 r/min摇床培养48 h,可达最大酶产量(37.6±0.8)mmol.min-1.L-1.
Strain CS 1-1 was isolated from the rich oil sample of Yuelu mountain and Xiangjiang river in Changsha city,which was identified as Aspergillus niger based on 18S rDNA sequence analysis.The fermentation conditions for lipase production by strain CS 1-1 were optimized,and the maximum lipase production of(37.6±0.8) mmol·min·L-1 was obtained at 28 ℃,180 r/min and pH 6.5 after 48 h Incubation using olive oil(1%) and sucrose(5 g/L) as combination of peptone and carbon sources,yeast extract(40 g/L) and ammonium sulfate(1 g/L) as nitrogen sources.
