摘要:对于不可逆电穿孔(irreversible electroporation,IRE)技术等治疗肿瘤的物理疗法而言,术后残存肿瘤细胞的粘附、侵袭和迁移能力能否被抑制,是关系到能否降低肿瘤复发和转移风险、巩固和保证治疗效果的重要问题。为探讨采用不可逆电穿孔治疗肿瘤时,所施加的μs脉冲电场对残存肿瘤细胞粘附、侵袭和迁移能力的抑制作用,以人宫颈癌细胞系Hela为对象,采用固定脉宽100/μs、频率1Hz、脉冲个数8个,电场强度分别为500、1000和1500V/cm的脉冲电场进行处理,通过基质胶的粘附实验、细胞体外侵袭和体外迁移实验,研究μs脉冲电场对肿瘤细胞上述3种能力的影响。研究表明,μs脉冲电场对Hela细胞的粘附能力、侵袭能力和迁移能力均有很好的抑制作用,且脉冲电场的场强越大,抑制效果越明显。研究证明了μs脉冲电场对抑制肿瘤复发和转移能力的作用,为进一步证实不可逆电穿孔治疗技术的优势及下一步的临床推广提供了新的实验依据。
It is of great importance for physical tumor therapies, including the irreversible electroporation (IRE), to inhibit the migratiov; adhesion and invasion abilities of the residual cancer cells after treatment. Whether or not the inhibition can be achieved determines' to a large extent, the therapeutic effects and the success to reduce the risk of cancer recurrence and metastasis. We investigate the effects of microsecond pulsed electric field (μsPEF) on cell migration, adhesion and invasion abilities with adhesion experiment, invasion and migration assays on human Hela cells. It is proved that/ISPEF can inhibit the three abilities above to a certain degree, and that the larger the.field strength applied is, the better inhibition can be resulted in. The experimental results demonstrate that t~sPEF has a good performance incancer recurrence and metastasis, which may serve to highlight the advantages of IRE for cancer treatment and promote its clinical application further.
