目的探索应用CpG寡脱氧核苷酸刺激对慢性淋巴细胞白血病(CLL)细胞染色体分析的影响。方法对70例CLL患者的外周血(或骨髓)标本,分别应用植物血凝素、CpG寡脱氧核苷酸和CpG寡脱氧核苷酸联合IL-2刺激培养72h,常规收获细胞制备染色体并进行R显带分析。应用组合探针进行间期细胞荧光原位杂交检测。结果植物血凝素组、CpG寡脱氧核苷酸组和CpG寡脱氧核苷酸联合IL-2组染色体分析成功率(可分析分裂象≥20个)分别为90.0%、68.6%和68.6%,异常核型检出率分别为3.2%、43.6%和43.6%,组合探针荧光原位杂交异常核型检出率为64.3%。结论应用CpG寡脱氧核苷酸刺激培养CLL细胞,可提高染色体异常检出率。
Objective To explore the effect of CpG-oligodeoxynucleotides (ODN) in chromosome study of chronic lymphocytic leukemia (CLL). Methods Blood or bone marrow cells of 70 CLL patients were cultured for 72 h with PHA , CpG-ODN and CpG-ODN combined with IL-2, respectively. Routine karyotype analysis with R banding technique and interphase fluorescence in situ hybridization (FISH) were performed. Results The metaphase number ≥ 20 was considered as successful stimulation, which in PHA, CpG-ODN and CpG-ODN combined IL-2 groups were 90.0% , 68.6% and 68.6%, respectively, and the detection rates of chromosome aberrations were 3.2% , 43.6% and 43.6% , respectively. The aberrations rates detected by interphase FISH with a panel of probes was 64.3%. Conclusion CpG-ODN DSP30 can effectively raise the detection rate of chromosome aberrations in CLL patients.
