测定了龙眼胚性愈伤组织体胚发生过程中若干生物大分子的动态变化.结果表明:龙眼胚性愈伤组织体胚发生过程中淀粉有2个累积峰值,第1次出现在胚性愈伤组织阶段,第2次出现在球形胚阶段,在鱼雷形胚阶段又有一个小回升,这时淀粉积累较心形胚和子叶形胚阶段高一些;随着胚胎的发育,DNA、RNA、总核酸含量的变化趋势大致相同,最高点均出现在心形胚阶段,其中RNA含量的变化较DNA、总核酸含量明显,蛋白质含量的变化曲线呈“S”形.这些生物大分子的动态变化表明,它们对于体胚的发生和发育起着重要作用。
The dynamic changes of some biomacromolecules were measured in the process of somatic embryogenesis from embryogenic callus in longan ( Dimocarpus longan Lour. ). There were 2 peaks of starch accumulation in the process of somatic embryogenesis, the first time was at the stage of embryogenic callus, the second time was at the stage of globular embryoids; and a small raise appeared at the stage of torpedo-shaped embryoids, and the content of starch was slightly higher than those of heart-shaped embryoids and cntyledonary embryoids. With the development of embryos, the tendency of the changes of the content of DNA, RNA and the total nucleic acids was approximately similar, and the peak values were all at the stage of cotyledonary embryoids, and the change of the content of RNA was more remarkable than those of DNA and the total nucleic acids. The tendency of change of the content of proteins almost took on the S-shaped curve. The dynamic changes of those biomacromolecules suggested that they could play an important role in somatic embryogenesis.
