半乳糖凝聚素-3(galectin-3,Gal-3)是一种多功能蛋白,参与细胞增殖、分化、凋亡、血管发生及肿瘤发生与转移等多种生命活动过程。前期工作表明一些两栖类动物存在个体水平的gal-3 cDNA分子多样性。鉴于哺乳动物尚缺乏这方面的研究,选择新西兰兔(Oryctolagus cuniculus)为实验材料,制备3只不同个体的肝脏第一链cDNA,通过DNA聚合酶链式反应扩增gal-3开放阅读框(open reading frame,ORF)并进行克隆测序。结果表明,兔gal-3 ORF中存在5个单核苷酸多态性(single nucleotide polymorphism,SNP)位点,导致11个不同的ORF,其中1个与Gen Bank注册的序列完全相同,另外10个均为本研究首次发现。从1~3号个体的肝脏分别获得6、6、2条不同的兔gal-3 ORF,揭示兔肝脏存在个体水平的gal-3 cDNA分子多样性。
Galectin-3 (Gal-3) is a multi-functional protein involved in many biological processes such as cell proliferation, differentiation, apoptosis, angiogenesis, oneogenesis and metastasis. In previous studies, gal-3 cDNA diversity was indicated in some amphibian species. To clarify if such diversity exists in mammals, the cloning and sequencing of gal-3 open reading frame (ORF) was performed from 3 individuals' liver first strand cDNAs of New Zealand rabbit Oryctolagus cuniculus. As the result, 5 single nucleotide polymorphism (SNP) were observed resulting in 11 different clones, one of which is the same as that deposited in GenBank, while the others are new and have never been reported. In fact, 6 different clones were obtained from both the first and the second individuals, and 2 were ohtained from the third one. All these analysis indicated ga/-3 cDNA diversity at individual level in the liver of O. cuniculus.
