目的分析黑龙江省尖锐湿疣组织中感染的人乳头瘤病毒(HPV)基因型分布情况,比较保守引物和序列特异引物PCR扩增方法。方法提取26例尖锐湿疣组织标本中DNA,用HPV L1区通用引物及HPV 6b、HPV11、HPV16、HPV18型L1区特异引物进行PCR扩增,分析尖锐湿疣组织中感染的HPV型别分布并比较两种扩增方法的结果。结果26例标本中,HPV 6b和11型感染例数分别为4和11例,各占15.4%和65.4%;HPV 16型感染2例,占总例数7.7%;有2例合并HPV6b/11型混合感染,占7.7%;1例合并HPV11/16/18型的混合感染,占3.8%。用通用引物扩增后26例标本中1例为阴性。结论黑龙江省地区尖锐湿疣组织HPV DNA检出率为100%,以低危型别11和6b型为主,偶见高危型别16、18型感染。鉴定HPV感染保守引物与型别特异性引物PCR方法相比,前者可导致少量假阴性产生。
Objective To analyze the genotypes of human papillomavirus(HPV) in acuminate condyloma biopsies in Heilongjiang province and compare of consensus and type-specific PCR primers for HPV L1 gene amplification.Methods The tissue DNA was extracted from 26 biopsy specimens of acuminate condyloma.The L1 gene of HPV was amplified with consensus primers followed by type-specific primers for HPV6b,HPV11,HPV16 and HPV18.Results HPV 6b and HPV 11 were the most common types,4 cases(15.4%) and 11 cases(65.4%),respectively;two were type 16(7.7%);two were HPV 6b and HPV11 overlapped(7.7%);one biopsy was HPV 11,HPV16,and HPV18 overlapped(3.8%).It indicated the local type of HPV 11 represented the epidemic strain in Heilongjiang province.PCR with consensus primers failed to detect one case(3.8%) of HPV infection that was positive by PCR detection with the type-specific primers.Conclusion All of these acuminate condyloma biopsies are HPV positive.The most common types in acuminate condyloma are HPV11 and HPV6b,whereas the high risk types HPV16 and HPV18 are also found.The sensitivity of PCR with the typespecific primers is higher than that of PCR with consensus primers.
