目的建立E-cadherin诱导的卵巢上皮细胞早期恶变模型,并对其生长增殖等生物学行为进行初步研究。方法采用含人E-cadherin基因CDH1全长cDNA的质粒转染中国仓鼠卵巢(Chinese hamster ovary,CHO)细胞,利用G418筛选稳定转染细胞株,通过Western Blot验证其E-cadherin的表达,然后通过CCK8细胞增殖实验及集落形成实验检测其生物学行为的变化。结果 CHO细胞转染CDH1基因后,高度表达E-cadherin蛋白,生长增殖水平显著提高,并具备了恶性细胞特有的非锚定依赖性生长的能力。结论构建了E-cadherin诱导的卵巢上皮细胞恶变模型,为卵巢肿瘤发生机制的研究提供了可靠的工具。
Objective To construct an E-cadherin induced cancer model from ovarian epithelial cells and do primary research on its biological behaviors such as proliferation and growth.Methods Chinese hamster ovary(CHO)cells were transfected by plasmid including whole human E-cadherin cDNA and stably screened with G418.Expression of E-cadherin was identified by Western Blot,and cell proliferation test with CCK8 and colony formation test were used to detect their possible malignant transformation of biological behaviors.Results A high level of E-cadherin expression,high ability of proliferation and acquisition of anchorage-independent growth were found in CHO cells transfected by the E-cadherin plasmid.Conclusion An E-cadherin induced cancer model of ovarian epithelial cells was successfully constructed to provide a valuable tool to study the mechanisms of ovarian cancer.
