目的探讨急性心肌梗死(AMI)后梗死心肌组织问质细胞衍生因子-1α(SDF-1α)表达水平的变化。方法雄性Wistar大鼠31只,随机分为AMI组16只和假手术组15只。AMI组采用开胸并结扎左冠状动脉前降支的方法建立AMI模型。假手术组仅开胸并穿结扎线,不结扎前降支。在AMI模型建立后6h、24h以及6d分别处死一批大鼠(每组每个时间点3-4只)。实时定量PCR法测定梗死区组织SDF-1α mRNA表达水平,免疫印迹法测定SDF-1α蛋白的组织表达水平。结果与假手术组相比,AMI组大鼠在梗死6h时SDF-1α mRNA表达水平明显升高,24h升高达到高峰(分别是假手术组的3.2倍和5倍,P〈0.05),6d表达水平下降。蛋白表达水平在6h处于峰值水平,随后即开始下降。结论AMI时局部心肌组织SDF-1α的表达呈动态变化,但SDF-1α蛋白表达水平与mRNA表达变化并不一致,可能与急性炎症期其他外源性物质释放(如血小板源性)有关。
Objective To investigate the rule of stromal cell-derived iactor-1α (SDF-1α) expression m mtarct area In experimental myocardial infarction. Methods Male Wistar rats were randomized into AMI group and sham group. AMI group was subjected ligation of coronary artery. Sham group did not underwent occlusion of coronary artery. 3-4 rats were sacrificed in 6 h, 24 h and 6 d following myocardial infarction. Real-time PCR was used to analyze the quantity of SDF-1α mRNA. The protein levels of SDF-Ict were detected by western blotting. Results Compared with sham group, SDF-1α mRNA levels in AMI group were increased at 6 h, reached to peak at 24 h(P 〈0.05), eventually decreased at 6 d. But SDF-1α protein level reached to peak at 6 h, and then began to decline. Conclusions The discrepancy between mRNA level and protein level of SDF-1α in infarcted area of AMI patients may be related to the release of other exogenous sub- stances ( eg. platelet) in the stage of acute inflammation.
