目的采用HPLC-CAD法研究巴戟天Morindae Officinalis Radix炮制前后寡糖类成分的HPLC—CAD指纹图谱变化,为科学评价和控制其质量提供可靠依据。方法采用Shodex Asahipak NH2(250mm×4.6mm,5gm)色谱柱,乙腈和水为流动相,梯度洗脱,柱温为25℃,进样量为10μL。CAD参数:气压241.36kPa,量程100pA。结果巴戟天不同炮制品的HPLC指纹图谱共有峰特征明显,不同炮制品指纹图谱有差异。炮制后峰面积显著升高,表明巴戟天经炮制后,寡糖的量明显增加,炮制品共确定了10个共有峰。结论建立了巴戟天寡糖类成分的HPLC特征图谱检测方法,可用于巴戟天及其炮制品的质量控制。
Objective To establish a sensitive and specific HPLC-CAD method which could provide a reliable evidence for the scientific evaluation and quality control ofMorindae Officinalis Radix (MOR). Methods Shodex Asahipak NH2 column (250 mm × 4.6 mm, 5 μm) was used at column temperature of 25 ℃. The mobile phase was composed of acetonitrile-water by gradient elution. The injection volume was 10 μL. The CAD parameters were pressure of 241.36 kPa and range of 100 pA. Results The characteristic of common peaks for fingerprints of different processed products of MOR is distinct, but there are differences among various processed products of MOR. The peak area was enlarged after processing which indicated the obvious increase of oligosaccharides and determination of ten common peaks. Conclusion The established HPLC fingerprint method is accurate and reliable, and has a good reproducibility and precision, which could be used for the quality control of MoR and its processed products.
