[目的]为了进行甘蔗多糖的深入研究和利用。[方法]对甘蔗叶多糖提取与含量测定进行研究。建立80%乙醇去杂-热水提取-乙醇沉淀-去色素的提取工艺,从甘蔗叶中提取粗多糖,用苯酚-硫酸法比色测定多糖的含量,在波长490nm处测定吸光值。[结果]结果表明,甘蔗多糖在80℃、95%乙醇沉淀的条件下多糖提取率最高,为1.34%,在8~80μg范围内具有良好的线性关系,平均回收率为97.9%,RSD=0.6%。[结论]此方法简便、准确率高、重现性好,可用于甘蔗多糖提取与含量测定。
[ Objective ]The research aimed to study and utilize polysaccharide in sugarcane deeply. [ Method] The extraction and content determination of polysaccharide in sugarcane leaves were studied. The extraction technology of removing impurity with 80% ethanol, extracting with hot water, precipitating with ethanol and getting rid of pigments was established. Polysaccharide was extracted from sugarcane leaves, and its content was determined by phenol- sulphuric acid method and the absorption value was measured at the wavelength of 490 nm. [ Result ] Extracting polysaccharide from sugarcane at the te perature of 80 ℃ and with 95% ethanol precipitation could get highest extraction rate of polysaccharide (being 1.34% ) with the average recovery of 97.9% and RSD of 0.6%. [ Conclusion] This method was simple with high accuracy and good repeatability, and could be used for extraction and content determination of polysaccharide in sugarcane.
