目的乙肝表面抗体是一种保护性抗体,利用基因重组法构建含乙肝表面抗原抗体Fab基因的重组腺相关病毒(rAAV-Fab),通过转染小鼠腿部骨骼肌,观察此种转染途径,Fab的表达情况。方法肌肉注射10^11、5×10^10、5×10^9 3种滴度的rAAV-Fab,感染小鼠腿部肌肉组织,用荧光免疫吸附法检测血清中Fab含量变化。免疫组化分析表达分布。病理组织学检测转染后组织损伤。结果小鼠肌注rAAV-Fab后,2个月内在血清中均有持续较高表达Fab,其后呈现较低趋势,直至第4个月观察结束。转染后,全身多个脏器组织切片均可检测到阳性信号,但3种病毒滴度转染比较,肝脏转染并无差异。主要脏器病理检查结果为阴性。结论骨骼肌转染rAAV-Fab能够有效介导Fab基因在小鼠体内表达,转染广泛,且此方法安全有效,为进一步实验研究提供了依据。
Objective To detect the expression of the recombinant adeno-associated vires containing Fab gene (rAAV-Fab) in mouse muscular tissue transfected via intramuscular injection. Methods rAAV-Fab with three different titers of 10^11 , 5 × 10^10, 5 × 10^9 were transfected to mouse musclar tissue. The changes of Fab level in serum were detected by Fluorescent immunoabsorption. The immunohistochemistry of tissue section displayed the distribution of Fab expression in vivo. The immunity damage induced by rAAV -Fab was detected by pathology analysis. Results The Fab was found in serum. The quantity of Fab waved with time and the rAAV-Fab titer. Fab expressed highly in two months after infection with rAAV-Fab, then decreased in the fourth month. Fab was detected in all over the body of mice in the three groups, especially in muscular tissue. The liver tissue sections after infection with the virus of three titers were compared and no different was found. Pathology analysis shown no damage in all organ tissue sections. Conclusion The rAAV -Fab can efficiently mediate Fab gene expression in mouse via intramuscular injection, which provide an safety and good method for further research.
