三肽囊素(Bursin)模拟肽阳性噬菌体对新城疫疫苗免疫效果的影响

研究了两个噬菌体展示三肽囊素模拟肽阳性噬菌体的活性。将100羽1日龄罗曼鸡随机分成5组(Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ组),每组20羽,在2日龄时,Ⅰ组注射4号阳性噬菌体(滴度为211),Ⅱ组注射6号阳性噬菌体(滴度为211),Ⅲ组注射6号阳性噬菌体(滴度为29),Ⅳ组注射一定剂量的Bursin,Ⅴ组为TBS对照组,在8日龄时接种新城疫(ND)Ⅳ系苗,以后每隔1周采用微量血凝抑制试验和MTT法测定各组的免疫效应。结果表明:阳性克隆(4号和6号)与Bursin在动物试验上具有相似的免疫增强作用,能提高体液免疫水平,对细胞免疫也有提高,且这种促进作用的大小与所用噬菌体滴度有关。

英文摘要:

The activity of the two phage - displayed Bursin - mimic peptide positive phages has been studied. 100 one -day -old Luoman chickens were divided into 5 groups randomly,and each group had 20 chickens. At the age of 2 days, group Ⅰ was the number 4 positive phage (2^11PFU) ,group Ⅱ the number 6 positive phage (2HPFU) , group Ⅲ the number 6 positive phage ( 2^9 PFU), group Ⅳ the Bursin group and group Ⅴ the TBS control group. They were all immunized by NDV live vaccine at the age of 8 days. Then, each week, Cellular immune function was studied with proliferation of ehook lymphocyte ( MTT assay) and the growth of lymphocyte was photomierographed, meanwhile,through micro - agglutinate plate agglutination,the serum antibodies of anti - Newcastle Disease were studied. The results showed as follows : both the positive clones ( number 4 and number 6) and Bursin have analogously immunological enhancement property. They can not only enhance and keep the level of serum antibody against NDV, but also have slightly strong promoting effect on cellular immune function, and the enhancement property of positive phage has correlation with their injeeion PFU.
