液相色谱多模式混合固定相同时具有多种分离机理,被越来越多的应用于复杂样品的分析。通过改变两种改性基团的投料比,制备了一组经典的RP/SCX双模式混合固定相,并对其进行了系统的评价研究。采用元素分析、离子交换当量测试,得到了-C18与-SO3H两种基团的比例,验证了空间位阻对-C18键合率的影响;分别在反相和离子交换模式下,考察了疏水性样品、离子样品的保留行为;并成功分离了含有6种不同类型化合物的混合物。评价结果表明,制备的固定相同时具有反相和离子交换色谱的双重保留机理,为此类固定相分离复杂样品提供了参考数据。
Mixed-mode stationary phases of HPLC had been applied to analyses of complicated samples more and more because of their multiple separation mechanisms.A number of classical RP/SCX mixed-mode stationary phases were prepared by changing the feed ratio of modifying agents.Then they were evaluated and researched systematically.The ratios of-C18 to-SO3 H were acquired by element analyses and ion exchange capacity tests which confirmed that the bond rates were affected by steric hindrance.The retention behaviors of hydrophobic samples and ion samples were studied under RP and IEC mode respectively.A mixture of six kinds of compounds was separated successfully.Results showed that the prepared stationary phase had double retention mechanisms of RP and IEC which offered references for this kind of stationary phase to separate complicated samples.
