目的:探讨细胞生长因子(GF)对生物电场(Ⅱ-诱导大鼠主动脉血管平滑肌细胞(VSMC)定向迁移的影响。方法:体外培养大鼠主动脉VCGMC,在不同条件培养基中用150mV·mm^-1 EF干预6h,显微成像及图象分析技术检测平滑肌细胞迁移情况。结果:在含有10%胎生血清(FBS)的培养基中,VSMC在EF作用下向阴极迁移。在无血清的培养基中,细胞没有定向迁移。在无血清培养基中补充血小扳源生长因子-BB、碱性成纤维细胞生长因子、转化生长因子-β1(PDGF-BB、bFGF、TGF-β1)时,部分恢复细胞向阴极迁移。结论:EF诱导的VSMC定向迁移依赖于血清的存在,血清中的生长因子与EF协同作用,促进细胞向阴极迁移。
Objective:To investigate the effects of growth factor(GF) on directed migration of cultrued vascular smooth muscle cells(VSMC) among electrical field Methods:Cultured VSMC were exposed to physiological electrical field 150mV/min for 6 hours in the presence of DMEM,DMEM+10% foctal bovine serum(FBS) or DMEM+GF,theinterval photographs of VSMC were taken and analyzed.Results:In media containing 10% FBS,these cells migrated cathodally.The cells cultured in serum-free medium showed no directed migration.All of platelet derived growth factor-BB(PDGF-BB),basic firbroblast growth factor(BFGF) and transforming growth factor-β1(TGF-β1) promoted VSMC directed migration in serum-free medium.Conclusion:Physiological electrical field induce VSMC directed migration.Some substances in serum are necessary to VSMC directed migration.including growth factors.
