为了解蛋鸡中禽白血病当前流行毒株类型及其gp85基因分子特征,在临床症状和病理组织学观察的基础上,采用ELISA抗原检测、特异性PCR以及亚群特异性免疫荧光试验(IFA),证实从黑龙江省某鸡场送检蛋鸡中分离到1株J亚群禽白血病病毒(ALV-J),命名为HLJ09 MDJ-1;克隆了其gp85基因,并进行了序列分析比较。结果表明,该分离株的gp85基因与国内外其他ALV-J毒株的核苷酸同源性为88.2%~98.2%。其中,HLJ09 MDJ-1与英国原型株HPRS-103的同源性最高(98.2%),与美国株ADOL-7501的同源性最低(88.2%),表明HLJ09 MDJ-1的gp85基因与HPRS-103有较近的亲缘关系。本研究从蛋鸡中分离到ALV-J毒株,为研究ALV-J宿主嗜性改变和ALV-J对蛋鸡致病机制提供了素材。
To find out the current prevalent ALV strain in layers and its molecular characteristics,a strain of subgroup J avian leukosis virus(ALV-J),named HLJ09MDJ-1,was isolated and identified by clinical symptoms and histopathological observation,PCR amplication and immunofluorescence assay special to ALV-J from layers suspected with ALVs infection in a farm of Helongjiang province in China.Moreover,the gp85 gene of HLJ09MDJ-1 was cloned and analyzed with other ALV-J strains published in GenBank.The results showed that the gp85 gene of HLJ09MDJ-1 shared 88.2%-98.2% homology in the level of nucleotide acid sequences with other ALV-J strains,among which,the highest homology(98.2%) with HPRS-103 and the lowest homology(88.2%) with ADOL-7501.Overall,these data suggest a close relativity between the gp85 genes of HLJ09MDJ-1 and HPRS-103.In conclusion,the HLJ09MDJ-1 strain isolated from egg-type chickens may contribute to the researches on the changes of ALV-J host range and pathogenicity to egg-type chickens.
